ARIAD (ARIA) and Merck decided to team up to develop an experimental cancer treatment, “AP23573″ - a new class of cancer small molecules that block mTOR and interfere with growth of cancer cells by starving them. The partnership could be worth more than $1 billion to Ariad, which has been testing the medication. How the market reacted is not important. The important news is that Merck has recognized the fact that mTOR is a credible target and AP23573 is doing the job.

Late-stage clinical development will start soon with the drug targeting metastatic sarcomas. The drug is also being tested for solid tumors and blood-based cancers.

Ariad will receive an initial payment of $75 million and up to $452 million more based on whether certain development goals are met. The deal could reach hundreds of millions in payments in milestone accomplishments, approval and meeting sales’ targets. Ariad will share profit with Merck in the United States and take royalties on the drug sales overseas.

In the news, Amgen and Daiichi Sankyo Company, Limited signed a collaboration and license agreement for the development and commercialization of denosumab in Japan. Denosumab is a monoclonal antibody that targets RANK Ligand (an essential mediator of cells that break down bone).

Denosumab, Amgen’s antibody is in clinical trials for the prevention and treatment of a broad range of bone loss conditions including osteoporosis, bone metastases, treatment-induced bone loss, multiple myeloma and bone erosions in rheumatoid arthritis. Amgen has granted Daiichi Sankyo exclusive rights to develop and commercialize denosumab in Japan in post-menopausal osteoporosis and oncology with the potential for additional indications. Amgen will receive exclusive worldwide rights to certain Daiichi Sankyo intellectual property to the extent applicable to denosumab. The financial terms include:

- An upfront payment to Amgen of $20 million.

- Daiichi Sankyo will pay royalties on annual net sales of denosumab in Japan in amounts commensurate with a major late stage product for the Japan market.

Daiichi Sankyo believes that denosumab has the potential to be a first-in-class, leading product in Japan for multiple indications. Denosumab’s potential applicability in oncology makes it an important part of the foundation for the firm’s growing oncology business.

Daiichi Sankyo has a full range of commercial capabilities, including in the primary care and hospital settings, a track record of successful large, first-in-class product launches and the financial strength to ensure appropriate investment in the product.

FDA APPROVES VECTIBIXâ„¢ FOR Metastatic Colorectal Cancer

Sept. 27, 2006–Amgen (AMGN) today announced that the FDA has approved Vectibixâ„¢ (panitumumab) following priority review. Vectibix is the first entirely human monoclonal antibody for the treatment of patients with epidermal growth factor receptor- (EGFr) expressing metastatic colorectal cancer after disease progression on, or following fluoropyrimidine-, oxaliplatin-, and irinotecan- containing chemotherapy regimens.

The FDA approval of Vectibix was based on a progression-free survival endpoint. Vectibix is the first anti-EGFr antibody shown to significantly improve progression-free survival in patients with metastatic colorectal cancer. Currently no data are available that demonstrate an improvement in disease-related symptoms or increased survival with Vectibix. Vectibix can be conveniently administered intravenously once every two weeks.

Vectibix is expected to be commercially available in early-to-mid October and will be priced at approximately 20 percent less than the other anti-EGFr antibody currently on the market.

“Vectibix is the first entirely human antibody for the treatment of colorectal cancer to be approved by the FDA. It provides another option for patients with metastatic colorectal cancer that have progressed on all available chemotherapy regimens,” said J. Randolph Hecht, M.D., director of the UCLA Gastrointestinal Oncology Program and clinical professor of Medicine, UCLA David Geffen School of Medicine, Los Angeles. “In a large, randomized clinical trial, Vectibix has been shown to delay progression of disease compared to best supportive care.”

Epidermal growth factor receptors are proteins that play an important role in cancer cell signaling. Vectibix is an entirely human IgG2 monoclonal antibody that binds with high affinity to EGF receptors. The goal of developing entirely human monoclonal antibodies is to offer effective targeted therapies with lessened risk of immune response against these agents.

“Our goal is to fulfill the promise of biotechnology to improve the way cancer is treated,” said Willard Dere, M.D., chief medical officer and senior vice president of Global Development at Amgen. “The approval of Vectibix allows us to build on our strong foundation in supportive care and move forward with our comprehensive approach to helping patients in their fight against cancer.”

“One out of 18 people in this country will develop colorectal cancer in their lifetime and 20 percent of colorectal cancers are found after the disease has spread to distant organs.” said Amy Kelly, director and co-founder of the Colon Cancer Alliance. “That means that a person in the U.S. is diagnosed with colorectal cancer every four minutes, heightening the need for new therapeutic options such as Vectibix.”

Marketing applications were simultaneously submitted to the European Medicines Agency (EMEA) in April 2006 and Health Canada, Australia and Switzerland in May 2006. AMGEN (AMGN) Results from a Phase 2 study presented at the 2006 European Society of Cardiology-Heart Failure (ESC-HF) have shown that, in patients with symptomatic heart failure and anemia, Aranesp® was well tolerated and effectively raised hemoglobin (Hb) (Abstract # 60734). Amgen also announced results from a prespecified pooled analysis including these data and data from a second Phase 2 study presented in March 2006 at the American College of Cardiology (ACC) Scientific Session showing that treatment with Aranesp may decrease the risk of heart failure hospitalization and all-cause mortality, and improve symptoms (Abstract #60731). These results require confirmation from a large Phase 3 randomized controlled clinical trial.
